The ISU Plant Transformation Facility offers multiple services related to crop genetic transformation. We are constantly working to improve our overall productivity and to consistently meet the needs of our customers.
Maize Genetic Transformation: Biolistic-mediatedBiolistic transformation is a physical way to deliver exogenous DNA into a cell. We use the BioRad PDS/He 1000 device and 0.6 µm gold particles for maize transformation. Our most frequently used maize genotype is Hi II (a hybrid derived from A188 and B73).

Maize Genetic Transformation: Agrobacterium-mediatedAgrobacterium tumefaciens is a soil bacterium that has the ability to transfer DNA into plant cells. We use disarmed Agrobacterium strains carrying standard binary vectors for transformation of immature embryos of maize Hi II genotype. Inbred maize (B104 genotype) is also offered using this protocol with sufficient lead time.

Soybean Genetic TransformationAgrobacterium-mediated transformation is used to transform this crop following a recently improved method that uses a half-seed explant from mature seed as the target tissue. Transformable soybean varieties include William 82 and Thorne.

Rice Genetic TransformationWe use our expertise in Agrobacterium-mediated transformation to transform rice callus cultures. Two transformable Japonica varieties include Nipponbare and Kitaake.

Transformation related servicesAlong with genetic transformation of crops our facility offers other transformation related services. You can purchase greenhouse grown maize ears, maize immature embryos, biolistic gun supplies and maize tissue culture media. In addition, we can introduce DNA construct into Agrobacterium strains for your transformation projects. Customers can even transform worms (C. elegans) using our biolistic gun.

Consulting and OtherOur experienced staff is here to assist you at every stage of plant transformation from construct design to greenhouse care of transgenic plants. We also provide non-transgenic tissue or seed upon request that can be used either as an experimental control or for laboratory/greenhouse practices.
